目的研究淫羊藿苷对氧糖剥夺PC12细胞所致炎症损伤的保护作用。方法采用PC12细胞复制氧糖剥夺2 h细胞损伤模型。实验分成6组,即正常组,模型组,尼莫地平组(10-5mol·L-1),淫羊藿苷高、中、低剂量组(10-5,10-6,10-7mol·L-1)。将淫羊藿苷各剂量组预给药1 h加氧糖剥夺期间给药2 h后,用MTT法检测细胞活力,ELISA法检测细胞上清液的肿瘤坏死因子-α(TNF-α)、白细胞介素-lβ(IL-1β)、白细胞介素-6(IL-6)含量。结果淫羊藿苷各剂量组均能显著降低氧糖剥夺PC12细胞上清液的TNF-α、IL-1β、IL-6含量,提高细胞的活力,与模型组比较,差异均有显著性意义(P<0.05,P<0.01)。结论淫羊藿苷能减轻氧糖剥夺PC12细胞所致的炎症损伤,具有细胞保护作用。
Objective To study the protective effect of icariin on inflammatory injury induced by oxygen glucose deprivation in PC12 cells. Methods PC12 cells were used to replicate the model of cell injury induced by oxygen deprivation for 2 h. The experiment was divided into 6 groups: normal group, model group, nimodipine group (10-5mol·L-1), icarrin high, medium and low dose group (10-5, 10-6, 10-7mol · L-1). Icariin pretreatment dose of 1 h plus oxygen deprivation for 2 h after administration, the cell viability was detected by MTT method, ELISA was used to detect the supernatant of tumor necrosis factor-α (TNF-α) Interleukin-1β (IL-1β), interleukin-6 (IL-6) levels were measured. Results Each dose of icariin could significantly decrease the levels of TNF-α, IL-1β and IL-6 in the supernatant of PC12 cells by oxygen-glucose deprivation and increase the viability of cells. Compared with the model group, the differences were significant (P <0.05, P <0.01). Conclusion Icariin can reduce the inflammatory injury induced by oxygen deprivation of PC12 cells and has the protective effect on cells.
