本研究的目的在于通过观察人的嗜硷性前躯细胞的变化,查清嗜硷性细胞产生的机制。患者为30岁,女,怀疑为慢性嗜硷细胞白血病,外周血白细胞10万/毫米~3,成熟嗜硷细胞占60%。取患者的外周血,肝素抗凝,用 Ficoll-泛影葡胺分离浓集有核细胞,作单层琼脂半固体培养,观察嗜硷细胞集落的形成。集落刺激因子用培养人胎盘的上清液。培养结束后,取出琼脂中之集落,放于玻片上干燥,甲苯胺蓝染色,染成具有粉红色至深紫色的细胞为嗜碱细胞。把20个嗜碱细胞以上的细胞团计为一个细胞集落,并把这种细胞
The purpose of this study is to investigate the mechanism of basophilic cell production by observing changes in human basophilic forequarters. The patient was 30 years old, female, suspected of chronic alkalophilic leukemia, peripheral leukocytes 100,000 / mm3, and 60% of mature basophils. Peripheral blood of patients was taken and anticoagulated with heparin. Nucleated cells were separated by Ficoll-diatrizoate and cultured in a single layer agar semi-solid to observe the formation of basophilic colonies. Colony-stimulating factor is used to culture human placenta supernatant. After the cultivation, the colonies in the agar were removed, dried on a glass slide, stained with toluidine blue, and stained with a pink to purple-colored cell as a basophilic cell.
