目的模拟肿瘤缺氧环境,观察缺氧诱导因子1-α(hypoxiainducible factor 1-α,HIF1-α)和生存素在结肠癌SW480细胞中的表达变化。方法用三气培养箱模拟肿瘤间断性缺氧环境,免疫荧光检测HIF1-α和生存素蛋白的表达部位,RT-PCR和Western blot检测HIF1-α和生存素mRNA及蛋白表达量的变化。结果 HIF1-α和生存素蛋白主要在结肠癌SW480细胞的细胞质中表达,其表达部位不受缺氧环境的影响。间断性缺氧状态下HIF1-α和生存素mRNA的表达量分别为0.53±0.01和0.40±0.03,明显高于常氧时的表达量(0.29±0.01和0.20±0.02),二者比较差异有统计学意义(P<0.01)。HIF1-α和生存素蛋白表达量在常氧状态下分别为0.25±0.01和0.23±0.01,间断性缺氧状态下分别为0.39±0.01和0.38±0.01,二者比较差异有统计学意义(P<0.01)。结论 HIF1-α和生存素蛋白主要在结肠癌SW480细胞的细胞质中表达,其表达部位不受间断性缺氧环境的影响;HIF1-α和生存素mRNA及蛋白的表达量在间断性缺氧的环境下明显增加。
Objective To investigate the hypoxia environment of tumor and to observe the expression of hypoxia induced factor 1-α (HIF1-α) and survivin in colon cancer SW480 cells. Methods HIF-1α and survivin were detected by immunofluorescence in three-atmosphere incubator. The expression of HIF1-α, survivin mRNA and protein were detected by RT-PCR and Western blot. Results HIF1-α and survivin were mainly expressed in the cytoplasm of SW480 colon cancer cells, and their expression sites were not affected by hypoxia. The expression of HIF1-α and survivin mRNA in intermittent hypoxia were 0.53 ± 0.01 and 0.40 ± 0.03, respectively, which were significantly higher than those in normoxia (0.29 ± 0.01 and 0.20 ± 0.02) Statistical significance (P <0.01). The expressions of HIF1-α and survivin were 0.25 ± 0.01 and 0.23 ± 0.01 respectively under normoxia and 0.39 ± 0.01 and 0.38 ± 0.01 respectively under intermittent hypoxia (P <0.05), the differences were statistically significant (P <0.01). Conclusions HIF1-α and survivin are mainly expressed in the cytoplasm of SW480 colon cancer cells, and their expression sites are not affected by the intermittent hypoxic environment. The expression of HIF1-α and survivin mRNA and protein in the intermittent anoxia Environment significantly increased.
