目的探讨galectin-3对骨髓间充质干细胞(MSCs)来源的内皮细胞增殖的影响。方法密度梯度离心法分离纯化SD大鼠骨髓MSCs,取第3代,加入血管内皮生长因子(VEGF)和碱性成纤维细胞生长因子(bFGF),体外诱导14 d,进行免疫组织荧光染色和电镜鉴定,用低、中、高不同浓度galec-tin-3(0.1μg/mL,1.0μg/mL和5.0μg/mL)对骨髓MSCs来源的内皮细胞作用24 h;采用MTT比色法观察不同浓度galectin-3对内皮细胞增殖的影响;利用流式细胞仪检测DNA细胞周期的变化。结果 (1)中高浓度galectin-3具有显著的促进血管内皮细胞增殖的作用(P<0.05);(2)流式细胞仪检测DNA细胞周期显示各组S期细胞明显增加(P<0.05),中高浓度galectin-3组G2M期细胞比例明显增加(P<0.05)。结论 galectin-3可以促进骨髓MSCs来源的内皮细胞增殖。
Objective To investigate the effect of galectin-3 on proliferation of endothelial cells derived from bone marrow mesenchymal stem cells (MSCs). Methods MSCs were isolated and purified from bone marrow of SD rats by density gradient centrifugation. The cells were passaged for 3 generations. VEGF and bFGF were added into the MSCs of SD rats for 14 days. The MSCs were stained with immunofluorescence staining and electron microscopy (0.1μg / mL, 1.0μg / mL and 5.0μg / mL) for 24 h. The MTT assay was used to observe the effect of different concentrations of galec-tin galectin-3 on the proliferation of endothelial cells; use of flow cytometry to detect DNA changes in cell cycle. Results (1) The high concentration of galectin-3 could significantly promote the proliferation of vascular endothelial cells (P <0.05). (2) The flow cytometry showed that the cell cycle of S phase in each group was significantly increased (P < The proportion of G2M phase cells in the medium and high concentration galectin-3 group increased significantly (P <0.05). Conclusion galectin-3 can promote the proliferation of bone marrow MSCs derived endothelial cells.
