目的提出利用具特殊活性乳酸菌来预防、治疗高尿酸血症方法,筛选具有较高降血尿酸活性的乳酸菌。方法基于RP-HPLC方法,通过检测培养液培养前后肌苷、鸟苷含量变化,筛选出具有最快核苷分解速率的乳酸菌株作为候选菌株。并将其施用于高尿酸血症模型大鼠,观察对模型大鼠血尿酸含量影响。结果获得具有最佳核苷分解速率的菌株DM9218。动物实验结果表明DM9218干预组的血尿酸浓度(219.25±21.98)μmol/L明显低于模型组(311.75±27.07)μmol/L(P<0.05)。结论菌株DM9218具有最快的核苷分解速率,对高尿酸血症大鼠具有明显的降血尿酸作用。菌株DM9218为植物乳杆菌(Lactobacillus plantarum)。
Objective To propose a method of preventing and treating hyperuricemia by using special active lactic acid bacteria and screening lactic acid bacteria with high activity of lowering blood uric acid. Methods Based on the RP-HPLC method, the lactic acid bacteria strains with the fastest rate of nucleoside degradation were screened out as candidate strains by detecting the content of inosine and guanosine before and after culture. The rats were administrated with hyperuricemia model rats to observe the effect on blood uric acid content in model rats. Results The strain DM9218 with the best nucleolytic rate was obtained. Animal experiments showed that serum uric acid concentration of DM9218 intervention group (219.25 ± 21.98) μmol / L was significantly lower than that of model group (311.75 ± 27.07) μmol / L (P <0.05). Conclusion The strain DM9218 has the fastest rate of nucleoside degradation and has a significant effect on lowering blood uric acid in hyperuricemic rats. Strain DM9218 is Lactobacillus plantarum.
