通过体外半固体琼脂培养分别观察PGF_(2α)对小鼠骨髓细胞CFU-GM增殖和骨髓细胞DNA合成的影响。测定结果表明,PGF_(2α)对CFU-GM及骨髓细胞具有双向调控作用。PGF_(2α)浓度在10~(-10)~10~(-8)mol/L时表现为刺激增殖作用,PGF_(aα)浓度在10~(-9)mol/L时刺激作用最强,集落形成比对照组多34.1%,~3H-TdR掺入率增加44.5%,而PGF_(2α)大于10~(-7)mol/L时则表现为抑制作用。集落形成与~3H-TdR掺入率的50%抑制率的PGF_(2α)浓度均接近于10~(-5)mol/L。同时,~3H-TdR掺入率的改变提示PGF_(2α)对CFU-GM增殖的调节是通过影响细胞内DNA合成来实现的。集落形态学分类结果表明,PGF_(2α)对粒系,巨噬系、粒单混合系三类型集落形成都表现为双向调控作用,而粒系对PGF_(2α)反应最为敏感。
The effects of PGF 2α on the proliferation of CFU-GM and the DNA synthesis of bone marrow cells were observed by semi-solid agar in vitro. The results showed that PGF_ (2α) had bidirectional regulation on CFU-GM and bone marrow cells. PGF_ (2α) showed a stimulative effect on proliferation when the concentration of PGF_ (2α) was between 10 ~ (-10) ~ 10 ~ (-8) mol / L and PGF_ (aα) The colony formation was 34.1% more than the control group, the incorporation rate of ~ 3H-TdR increased by 44.5%, while the inhibitory effect was observed when the PGF 2α concentration was greater than 10 -7 mol / L. The concentration of 50% inhibition of colony formation and 3H-TdR incorporation were all close to 10 -5 mol / L. At the same time, the change of ~ 3H-TdR incorporation rate indicated that the regulation of CFU-GM proliferation by PGF 2α was achieved by affecting DNA synthesis in cells. Colony morphological classification results showed that PGF_ (2α) showed bidirectional regulation on the formation of three types of colony, macrophage and monogamy lines, while the granulocyte line was most sensitive to PGF_ (2α).
