采用直接逆转录一多聚酶链反应(RT-PCR)对加标海水、海泥、毛蚶的浓缩样品,用不同消毒方法处理过的甲型肝炎病毒(HAV)及毛蚶实测样品进行了HAV-RNA分析。结果表明,该方法可检出加标海水、海泥及毛蚶中的HAV-RNA;对苯酚消毒灭活的HAV检验结果表明含有完整核酸序列的灭活病毒也能用RT-PCR方法检出;毛蚶实测样品结果表明,只有1988年上海毛蚶RT-PCR阳性,但细胞培养间接免疫荧光法检测结果为阴性,说明毛蚶冻存太久(≥5年),HAV已不具感染性。直接RT-PCR方法简便敏感,不需超速离心及其它特殊设备,在普通实验室也能使用。
HAV-RNA analysis was performed on samples of concentrated seawater, sea mud and hairy mildew using direct reverse transcription-polymerase chain reaction (RT-PCR), samples of hepatitis A virus (HAV) treated with different disinfection methods, . The results showed that this method could detect HAV-RNA in spiked seawater, sea mud and pomfret. HAV test showed that the inactivated virus containing the complete nucleic acid sequence could also be detected by RT-PCR. The results of real test samples showed that only RT-PCR was positive in 1988, but indirect immunofluorescence was negative in cell culture, indicating that it had been frozen for too long (≥5 years) and HAV was not infectious. The direct RT-PCR method is simple and sensitive, does not require ultracentrifugation and other special equipment, can also be used in ordinary laboratories.
