目的:探讨Th1/Treg细胞转录因子T-bet和FOXP3 mRNA及相关细胞因子IFN-γ﹑IL-6、TGF-β1在儿童重症肌无力(MG)中的变化,为临床开展新方法治疗儿童MG提供理论依据。方法:应用RT-PCR技术检测31例MG患儿和20例健康儿童外周血单个核细胞(PBMC)T-bet和FOXP3 mRNA的表达情况;应用ELISA法检测相关细胞因子IFN-γ、IL-6、TGF-β1的浓度变化。结果:与健康儿童比较,MG患儿T-bet和FOXP3 mRNA表达增高(P<0.05);T-bet mRNA与FOXP3 mRNA的表达比值,MG患儿高于健康儿童(P<0.05);MG患儿和健康儿童的IFN-γ、IL-6和TGF-β1浓度比较差异无统计学意义(P>0.05)。结论:T-bet和FOXP3 mRNA在MG患儿中表达异常增高,表明Th1/Treg细胞参与了MG的发病过程;MG患儿的IFN-γ、IL-6和TGF-β1浓度无明显异常,提示MG患儿免疫功能紊乱程度较轻。
Objective: To investigate the changes of Th1 / Treg cells transcription factor T-bet and FOXP3 mRNA and related cytokines IFN-γ, IL-6 and TGF-β1 in children with myasthenia gravis (MG) Provide a theoretical basis. Methods: The expression of T-bet and FOXP3 mRNA in peripheral blood mononuclear cells (PBMCs) of 31 children with MG and 20 healthy children were detected by RT-PCR. Results: Compared with healthy children, the expression of T-bet and FOXP3 mRNA in MG children increased (P <0.05); the expression ratio of T-bet mRNA and FOXP3 mRNA in MG children was higher than that in healthy children (P <0.05); MG patients There was no significant difference in the concentrations of IFN-γ, IL-6 and TGF-β1 between children and healthy children (P> 0.05). CONCLUSION: The abnormal expression of T-bet and FOXP3 mRNA in MG patients shows that Th1 / Treg cells are involved in the pathogenesis of MG. The concentrations of IFN-γ, IL-6 and TGF-β1 in MG patients were not significantly different MG children with mild immune dysfunction.
