利用RAPD标记对来自浙江杭州西湖、湖北利川、重庆石柱三大主产区的12份莼菜材料进行遗传多样性及亲缘关系分析.从50个RAPD随机引物中筛选出6个引物,对所有供试材料扩增共获得68条带,其中多态性条带24条,占35.3%,平均每个引物扩增11.3条带.用POPGENE 1.32软件进行数据分析,结果表明:莼菜在种水平的遗传多样性Ht为0.1752,Shannons信息指数Hs为0.2777;种群水平的遗传多样性Ht为0.1287,Shannons信息指数Hs为0.1896.说明莼菜的遗传多样性较低.大部分遗传变异存在于种群内(73.43%),种群间的基因流为1.3816.应用Neis遗传距离进行UPGMA聚类分析的结果显示,重庆产区3份莼菜材料首先与湖北产区的4份材料聚在一起,然后再与浙江产区的莼菜相聚.表明重庆产区莼菜和湖北产区莼菜的亲缘关系更近.
RAPD markers were used to analyze the genetic diversity and phylogenetic relationships of 12 Chinese cabbage materials from three main producing areas in West Lake in Hangzhou, Zhejiang Province, Lichuan, Chongqing and Shizhu, Zhejiang province. Six primers were screened from 50 RAPD random primers, A total of 68 bands were obtained, of which 24 bands were polymorphic, accounting for 35.3%, with an average of 11.3 bands per primer. The data analysis using POPGENE 1.32 software showed that the genetic diversity Ht was 0.1752, Shannon’s index Hs was 0.2777, Ht was 0.1287 and Shannon’s index Hs was 0.1896, which indicated that the genetic diversity of Brasenia schreberi was lower.Most of the genetic variation existed in the population (73.43%) and the gene flow among populations was 1.3816. According to UPGMA clustering analysis using Nei’s genetic distance, the results showed that the three materials from Chongqing were clustered together with four materials from Hubei Province before With the Zhejiang area of common vegetables together, indicating that Chongqing and Chongqing production of vegetables and water repellent products of Brachythecium closer relationship.
