目的观察治疗剂量环孢素A(CsA)、他克莫司(FK506)和雷帕霉素(Rapa)对独肾模型大鼠心肌酶谱及心脏组织病理的影响。方法将肾移植术后CsA、FK506及Rapa首剂治疗剂量按公式换算为大鼠的用药剂量。40只独肾模型SD大鼠随机分为正常组、对照组、CsA组25 mg/(kg·d)、FK506组0.8 mg/(kg·d)及Rapa组0.2 mg/(kg·d)),每组8只,持续给药8周后称取大鼠体重,检测CK、CK-MB、LDH、AST等心肌酶谱指标,并观察各组大鼠心脏组织的病理改变(HE染色)。结果 CsA组与其他组大鼠相比体重增长值明显降低(P<0.05);CsA组的CK、CK-MB、LDH、AST均明显高于其他组(P<0.05),而正常组、对照组、FK506组及Rapa组组间CK、CK-MB、LDH、AST均无明显差异(P>0.05);CsA组心肌有不同程度损伤,FK506组及Rapa组心肌组织基本正常。结论治疗剂量CsA对心肌产生不同程度的损害,但FK506及Rapa对心肌损害较小。
Objective To observe the effects of CsA, FK506 and Rapa on cardiac enzymes and histopathological changes in a rat model of one kidney. Methods The dose of CsA, FK506 and the first dose of Rapa after renal transplantation were converted into the dosage of rats according to the formula. 40 SD rats were randomly divided into normal group, control group, 25 mg / (kg · d) in CsA group, 0.8 mg / (kg · d) in FK506 group and 0.2 mg / (kg · d) in Rapa group) , 8 rats in each group. After 8 weeks of continuous administration, the body weight of rats was weighed and the indexes of CK, CK-MB, LDH, AST and other enzymes were measured and the pathological changes of heart tissues of each group were observed (HE staining). Results The weight gain of CsA group was significantly lower than that of other groups (P <0.05). The levels of CK, CK-MB, LDH and AST in CsA group were significantly higher than those in other groups (P <0.05) There was no significant difference in CK, CK-MB, LDH and AST between FK506 group and Rapa group (P> 0.05). The myocardial tissue of CsA group had different degrees of injury, and the myocardial tissues of FK506 group and Rapa group were normal. Conclusion The treatment of CsA has different degrees of myocardial damage, but FK506 and Rapa have less damage to the myocardium.
