目的检测异常黑胆质成熟剂中总黄酮和总酚的含量。方法以芦丁为对照品,采用NaNO2-Al(NO3)3-NaOH比色法测定了总黄酮含量;以没食子酸为对照品,采用Folin-Ciocalteu比色法测定了总酚含量。结果芦丁浓度在0.011~0.130 mg/ml范围内线性关系良好,最低检测限为0.06μg/ml,最低定量限为0.18μg/ml。该方法加样回收率为95.1%~97.5%,精密度为1.34%,水提物和醇提物中总黄酮含量分别为16.2 mg/g和19.0 mg/g。没食子酸浓度在1.1~7.7μg/ml范围内线性关系良好,最低检测限为0.01μg/ml,最低定量限为0.03μg/ml。该方法加样回收率为99.5%~104.8%,精密度为3.39%,水提物和醇提物中总酚含量分别为38.28 mg/g和21.54 mg/g。结论此两种方法灵敏、准确性强,精密度高,重复性好,分别可以用作异常黑胆质成熟剂中总黄酮和总酚含量的测定。
Objective To detect the content of total flavonoids and total phenols in abnormal savda mature agent. Methods The content of total flavonoids was determined by NaNO2-Al (NO3) 3-NaOH method with rutin as reference substance. The content of total phenols was determined by Folin-Ciocalteu colorimetric method using gallic acid as control. Results The linear range of rutin was 0.011 ~ 0.130 mg / ml with the lowest detection limit of 0.06 μg / ml and the lowest limit of quantification of 0.18 μg / ml. The recoveries of this method ranged from 95.1% to 97.5% with a precision of 1.34%. The total flavonoids content in water extracts and alcohol extracts were 16.2 mg / g and 19.0 mg / g, respectively. The linear range of the concentration of gallic acid in the range of 1.1 ~ 7.7μg / ml was good with the lowest detection limit of 0.01μg / ml and the lowest limit of quantification of 0.03μg / ml. The recoveries of this method were 99.5% -104.8% with the precision of 3.39%, and the total phenols contents in water extracts and alcohol extracts were 38.28 mg / g and 21.54 mg / g, respectively. Conclusion These two methods are sensitive, accurate, accurate and reproducible. They can be used as the determination of total flavonoids and total phenols in Abiotic Savda.
