目的研究酸性成纤维细胞生长因子(acidicfibroblastgrowthfactor,aFGF)对睫状体色素上皮(pigmentedepithelial,PE)细胞的影响。方法将体外培养的牛眼PE细胞置于含不同浓度aFGF的培养液内,用倒置相差显微镜观察细胞的形态结构,通过细胞计数及MTT比色分析测定细胞的增殖。结果在aFGF作用下,PE细胞形态良好、增殖旺盛,而以10ngml-1的浓度效力最佳。结论aFGF有助于在PE细胞体外培养中获得更多的实验材料,或在临床用以治疗房水分泌减少的眼科疾病。
Objective To investigate the effect of acidic fibroblast growth factor (aFGF) on pigmentedepithelial (PE) cells. Methods Bovine PE cells cultured in vitro were cultured in culture medium containing different concentrations of aFGF. The morphological changes of cells were observed by inverted phase contrast microscope. Cell proliferation was measured by cell counting and MTT assay. Results Under the action of aFGF, PE cells were well-formed and proliferated well, and the best concentration was 10ngml-1. CONCLUSIONS: aFGF can help to obtain more experimental materials in PE cell culture in vitro, or can be used clinically to treat ophthalmic diseases with reduced aqueous humor secretion.
