目的探讨抑癌基因 LRIG1对膀胱癌细胞 BIU87侵袭能力的影响。方法用脂质体介导的基因转染技术,将 pLRIG1-GFP 质粒转染人膀胱癌细胞 BIU87,用 G418筛选出抗性克隆。用Boyden 小室观察转染前后细胞侵袭能力的变化,并用逆转录-聚合酶链式反应(RT-PCR)法和Western-blot 检测转染前后 LRIG1和表皮生长因子受体(EGFR)的 mRNA 和蛋白表达水平。结果转染 pLRIG1-GFP 后的 BIU87细胞侵袭能力显著降低,LRIG1的 mRNA 和蛋白表达水平明显升高,而EGFR 的 mRNA 和蛋白表达水平则显著降低(P<0.01)。结论 LRIG1对膀胱癌细胞 BIU87的侵袭能力有显著的抑制作用。
Objective To investigate the effect of tumor suppressor gene LRIG1 on invasiveness of bladder cancer cell line BIU87. Methods The plasmid pLRIG1-GFP was transfected into human bladder cancer cell line BIU87 by liposome-mediated gene transfection. Resistant clones were screened by G418. The invasion of cells was observed with Boyden chamber before and after transfection. The mRNA and protein of LRIG1 and EGFR were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western-blot The expression level. Results The invasion ability of BIU87 cells transfected with pLRIG1-GFP was significantly decreased, the mRNA and protein expression of LRIG1 was significantly increased, while the mRNA and protein expression of EGFR were significantly decreased (P <0.01). Conclusion LRIG1 has a significant inhibitory effect on the invasiveness of bladder cancer cells BIU87.
