研究比较了鹅掌楸花粉的几种保存条件。结果表明:(1)新鲜花粉在超低温(-196℃,LN2)条件下可保持5d。花粉存活率(76.51%)与对照(新鲜花粉)几乎相同(75.55%)。在电镜下观察其超微结构无异常变化。而保存15d的花粉细胞超微结构发生严重损伤,内膜系统及细胞器降解。25d后几乎观察不到存活的花粉。(2)在-25℃保存条件下,以二甲基亚砜为保护剂,添加蔗糖作为辅佐剂保存效果较好。(3)4℃不宜作为鹅掌楸花粉的保存条件,5d后花粉的萌发率降至23.51%;而且大部分花粉细胞内膜系统严重损伤,15d后细胞质及核质“空泡化”,发生致死性破坏。
The study compared the conservation conditions of Liriodendron pollen. The results showed that: (1) Fresh pollen could be kept for 5 days under ultra-low temperature (-196 ℃, LN2). The pollen viability (76.51%) was almost the same as the control (fresh pollen) (75.55%). No abnormal changes in the ultrastructure were observed under electron microscope. The ultrastructure of pollen cells preserved for 15 days severely damaged, and the intima system and organelles were degraded. Almost no viable pollen was observed after 25 days. (2) Under the condition of -25 ℃ preservation, dimethylsulfoxide as the protective agent, the addition of sucrose as co-adjuvant is better. (3) 4 ℃ should not be used as the preservation condition of the pollen of Liriodendron chinense, the germination rate of pollen decreased to 23.51% after 5 days; and most of the pollen cell endomembrane system was severely damaged. After 15 days, the cytoplasm and nucleoplasm were “vacuolization” , Fatal damage occurred.
