考察人参中提取的寡糖粗提物(CGOS)在体外对小鼠巨噬细胞的激活作用。首先用水提的方法获得人参寡糖粗提物,并确定了最佳提取工艺。通过研究CGOS对巨噬细胞吞噬功能、分泌一氧化氮(NO)、活性氧(ROS)的水平、诱导型一氧化氮合酶(iNOS)活性的影响,研究其对巨噬细胞激活作用。结果显示,CGOS在10-100μg/ml能够增强巨噬细胞的吞噬功能,并呈现良好的剂量依赖关系;在该浓度范围内iNOS活性增强,导致NO的产量显著增加,最佳作用浓度为100μg/ml;CGOS在10~200μg/ml浓度范围促进ROS的释放。上述结果表明,CGOS对腹腔巨噬细胞有激活功能。
To investigate the activation of mouse macrophages in vitro by using extracted crude oligosaccharide (CGOS) from ginseng. Firstly, the crude extract of ginseng oligosaccharide was obtained by water extraction and the best extraction process was determined. The effects of CGOS on macrophage phagocytosis, the secretion of nitric oxide (NO), reactive oxygen species (ROS) and the activity of inducible nitric oxide synthase (iNOS) were studied to investigate its effect on macrophage activation. The results showed that CGOS at 10-100μg / ml could enhance the phagocytosis of macrophages in a dose-dependent manner. Increasing of iNOS activity resulted in a significant increase of NO production at the optimal concentration of 100μg / ml; CGOS promoted the release of ROS at the concentration range of 10 ~ 200μg / ml. The above results indicate that CGOS can activate peritoneal macrophages.
