目的:建立α-细辛脑原位凝胶含量测定和体外释放度测定方法。方法:采用HPLC法,SHIM-PACK VP-ODS色谱柱(4.6 mm×250 mm,5μm),流动相甲醇-水(75∶25),检测波长258 nm,采用磷酸盐缓冲溶液(p H 7.4)作为释放液。结果:含量测定中α-细辛脑在10.4~104 mg·L-1与峰面积具有良好的线性关系(r=1),平均回收率100.6%,RSD 2.2%。体外释放度试验中α-细辛脑在2.06~82.4 mg·L-1线性较好(r=0.999 9),平均回收率95.83%,RSD 2.5%。结论:建立的分析方法具有专属性好、准确和可靠的特点,可为后续研究奠定基础。
OBJECTIVE: To establish a method for the determination of α-asarone in situ gel and in vitro release. METHODS: The mobile phase consisted of methanol-water (75:25), SHIM-PACK VP-ODS column (4.6 mm × 250 mm, 5 μm) with detection wavelength of 258 nm and phosphate buffer solution (pH 7.4) As a release solution. Results: The content of α-asarone in the determination of 10.4 ~ 104 mg · L-1 peak area with a good linear relationship (r = 1), the average recovery rate of 100.6%, RSD 2.2%. In vitro release test showed that α-asarone had a good linearity (r = 0.999 9) at 2.06-82.4 mg · L-1 with an average recovery of 95.83% and RSD 2.5%. Conclusion: The established analytical method has the characteristics of good specificity, accuracy and reliability, which can lay the foundation for further research.
