目的研究川芎嗪对结肠炎模型小鼠血管细胞黏附分子-1(Vascular cell adhension molecule-1,VCAM-1)表达的影响,探讨川芎嗪在治疗溃疡性结肠炎(Ulcerative colitis,UC)中的作用。方法将小鼠随机分为正常对照组、模型组和川芎嗪组,采用葡聚糖硫酸钠(Dextran sulfate sodium,DSS)诱导小鼠UC模型,观察UC炎症评价指标[疾病活动指数(Disease activityindex,DAI)、结肠组织形态学及组织学损伤],采用免疫组化法检测VCAM-l在结肠黏膜中的表达。结果与正常对照组比较,模型组的炎症评价指标均明显增高(P<0.01);正常对照组结肠黏膜中几乎不表达VCAM-1,模型组VCAM-1的表达明显升高(P<0.01),而川芎嗪组VCAM-1的表达较模型组明显降低(P<0.01)。结论 DSS诱导的结肠炎模型小鼠中VCAM-1表达增强,川芎嗪具有抑制VCAM-1表达的作用。
Objective To investigate the effect of ligustrazine on the expression of vascular cell adhesion molecule-1 (VCAM-1) in mice with colitis and explore the role of ligustrazine in the treatment of ulcerative colitis . Methods Mice were randomly divided into normal control group, model group and ligustrazine group. Dextran sulfate sodium (DSS) was used to induce UC model in mice. The inflammation index [disease activity index DAI), colon histomorphology and histological damage]. The expression of VCAM-1 in colonic mucosa was detected by immunohistochemistry. Results Compared with the normal control group, the indexes of inflammatory evaluation in model group were significantly increased (P <0.01). VCAM-1 expression in colonic mucosa was almost not observed in normal control group, and the expression of VCAM-1 in model group was significantly increased (P <0.01) , While the expression of VCAM-1 in ligustrazine group was significantly lower than that in model group (P <0.01). Conclusions The expression of VCAM-1 in DSS-induced colitis model mice is enhanced, and ligustrazine can inhibit the expression of VCAM-1.
