本研究探索白血病人的骨髓间充质干细胞是否也具有白血病恶变特征。分别分离培养急性早幼粒细胞白血病(APL)及慢性髓系白血病(CML)患者的骨髓间充质干细胞并与正常对照比较其形态、生长曲线、细胞表面标志及是否带有白血病细胞的恶性克隆性的基因异常标志。结果表明:白血病患者的骨髓间充质干细胞在细胞生物学特征方面与正常健康人无任何明显差异,未发现白血病患者的骨髓间充质干细胞带有白血病细胞的恶性克隆性基因改变。结论:来源于白血病患者的骨髓间充质干细胞并无相应白血病细胞的克隆性异常,提示可能作为自体移植的辅助治疗和细胞治疗或基因治疗的可供选择的细胞载体。
This study explored whether Leukemia human bone marrow mesenchymal stem cells also have the characteristics of leukemia malignant transformation. Bone marrow mesenchymal stem cells from patients with acute promyelocytic leukemia (APL) and chronic myeloid leukemia (CML) were isolated and compared with normal controls to compare their morphology, growth curve, cell surface markers and malignant clones with leukemia cells Sexual signs of abnormal gene. The results showed that in patients with leukemia, BMSCs had no significant difference in the biological characteristics of normal people, and no change of malignant clonogenic genes in BMSCs with Leukemia was found in leukemia patients. CONCLUSION: Bone marrow-derived mesenchymal stem cells derived from leukemia patients do not have the corresponding clonal abnormalities of corresponding leukemia cells, suggesting that they may serve as an alternative cell carrier for adjuvant therapy of autotransplantation and cell therapy or gene therapy.
