目的:观察祛痰除湿剂对原代培养骨关节炎(OA)患者的膝关节软骨细胞中Ⅱ型胶原、X型胶原和相关代谢酶MMP-1、MMP-3、MMP-13 mRNA表达的影响,探讨祛痰除湿剂治疗骨关节炎的作用及其机制。方法:原代培养骨关节炎患者的膝关节软骨细胞,用第三代细胞进行试验。关节软骨细胞经含药血清作用72h后,采用Real Time-PCR法检测细胞中Ⅱ型胶原(Col Ⅱ)、Ⅹ型胶原(Col X)、基质金属蛋白酶-1、基质金属蛋白酶-3(MMP-3)、基质金属蛋白酶-13的mRNA表达。结果:OA患者膝关节软骨细胞经祛痰除湿剂含药血清作用后Col Ⅱ mRNA表达明显高于仅使用胎牛血清培养的空白对照组,经10%祛痰除湿剂含药血清作用后Col X及MMP-1、MMP-3、MMP-13 mRNA表达均明显低于空白对照组。结论:祛痰除湿剂可调节OA患者关节软骨中胶原及相关金属蛋白酶(MMPs)的mRNA表达,从而对退变的软骨细胞有一定的修复作用并可延缓关节软骨细胞的进一步退变。
OBJECTIVE: To observe the effect of expectorant dehumidification agent on the expressions of type Ⅱ collagen, type X collagen and related metabolic enzymes MMP-13, MMP-13 mRNA in articular chondrocytes of primary osteoarthritis patients , To explore the role of expectorant dehumidification agent in the treatment of osteoarthritis and its mechanism. Methods: Primary cultured chondrocytes from patients with osteoarthritis were tested with the third generation of cells. The cartilage cells of the articular cartilage were treated with the serum for 72 hours. Real-time PCR was used to detect the expression of Col Ⅱ, Col X, MMP-1 and MMP- 3), mRNA expression of matrix metalloproteinase-13. Results: The expression of Col Ⅱ mRNA in knee joint chondrocytes of OA patients was significantly higher than that of the control group treated with serum containing expectorant and desiccant. Compared with the blank control group cultured with fetal bovine serum, Col X The mRNA expression of MMP-1, MMP-3 and MMP-13 were significantly lower than that of the blank control group. CONCLUSION: Expectorant desiccant can regulate the mRNA expression of collagen and related metalloproteinases (MMPs) in articular cartilage of patients with OA, which can repair degenerated chondrocytes and delay the further degeneration of articular chondrocytes.
