目的:观察脉冲强磁场联合姜黄素引起膀胱肿瘤细胞BIU-87凋亡的作用。方法:将体外培养的膀胱肿瘤细胞癌株BIU-87随机分为A、B和C 3组。A组为空白对照组,B、C组均经姜黄素处理,C组处理后置于脉冲强磁场中干预(8T,15HZ),每天2 h。分别于干预后的第1、2、3天时采用显微镜观察肿瘤细胞形态、MTT法检测肿瘤细胞生长抑制率。结果:与A组比较,肿瘤细胞显微镜下处理后的B、C组,均出现典型的凋亡细胞形态,以C组表现更加明显;MTT检测,B、C组肿瘤细胞生长抑制率明显上升(均P<0.01),且呈时间依赖关系,第1~3天肿瘤细胞增殖抑制率B组明显低于C组(18.6%、40.6%、51.4%与26.9%、50%、61.4%,P<0.01);结论:脉冲强磁场联合姜黄素可促进膀胱肿瘤细胞的凋亡和抑制其生长。
Objective: To observe the effect of pulsed magnetic field combined with curcumin on the apoptosis of bladder tumor cells BIU-87. Methods: Bladder cancer cell line BIU-87 cultured in vitro was randomly divided into A, B and C 3 groups. A group was blank control group, B, C group were treated with curcumin, C group placed in a pulse magnetic field intervention (8T, 15HZ), 2 h daily. The morphological changes of tumor cells were observed with microscope after 1, 2 and 3 days respectively. The growth inhibition rate of tumor cells was detected by MTT assay. Results: Compared with group A, typical apoptotic cell morphology appeared in group B and group C after treatment with tumor cells, which was more obvious in group C. The inhibitory rate of tumor growth in group B and C was significantly increased by MTT (P <0.01). The inhibitory rates of tumor cell proliferation on day 1 to 3 in group B were significantly lower than those in group C (18.6%, 40.6%, 51.4% and 26.9%, 50% and 61.4%, P < 0.01). CONCLUSION: Pulsed intense magnetic field combined with curcumin can promote the apoptosis and inhibit the growth of bladder tumor cells.
