目的观察白藜芦醇对2.5-己二酮(2.5-HD)所致睾丸生精障碍的生精恢复作用。方法40只雄性SD大鼠随机分为5组(n=8),A组正常喂饲,B、C、D、E组均饮用含1%2.5-HD的水5周,然后C、D、E组用不同浓度的白藜芦醇(分别为0.4%、0.8%和1.6%)进行治疗9周。比较各组外表体征、体重增加值、睾丸重量的变化,HE染色与免疫组化分析比较各组生精小管数目、直径以及生精细胞膜c-kit蛋白表达情况。结果2.5-HD导致大鼠体质瘦弱,皮肤松弛,毛色暗淡,体重增长减慢,睾丸萎缩等变化;形态学显示生精上皮萎缩,生精细胞发育停滞,c-kit蛋白表达受到抑制。经过白藜芦醇治疗后,所有大鼠外表体征得到恢复,接近正常,体重增加值和睾丸重量恢复增长更为明显。组织学显示睾丸生精恢复,c-kit蛋白重新获得表达,但与正常组比较仍有异常,生精小管数目、直径随白藜芦醇用量增加而更为接近正常。结论白藜芦醇能够改善2.5-HD所致睾丸生精障碍,促进大鼠生精恢复。
Objective To observe the effect of resveratrol on the spermatogenesis of testicular spermatogenic disorders caused by 2.5-HD. Methods Forty male Sprague-Dawley rats were randomly divided into 5 groups (n=8). Group A received normal feeding. B, C, D and E groups all received water containing 1% 2.5-HD for 5 weeks, then C, D, Group E was treated with different concentrations of resveratrol (0.4%, 0.8%, and 1.6%, respectively) for 9 weeks. The changes of external signs, body weight gain, and testicular weight were compared between groups. HE staining and immunohistochemical analysis were used to compare the number and diameter of spermatogenic tubules and the expression of c-kit protein in spermatogenic cell membranes. RESULTS: 2.5-HD resulted in thin, loose skin, dark coat, slow weight gain, and testicular atrophy. Morphology showed that the spermatogenic epithelium atrophied, the development of spermatogenic cells was arrested, and c-kit protein expression was inhibited. After treatment with resveratrol, all the external signs of the rats recovered and were close to normal, and the increase in weight gain and testicle weight increased more significantly. Histology showed that testicular spermatogenesis was restored and c-kit protein was re-expressed, but it was still abnormal compared with the normal group. The number and diameter of seminiferous tubules were closer to normal with the increase of resveratrol dosage. Conclusion Resveratrol can improve the disorder of spermatogenesis induced by 2.5-HD and promote the recovery of spermatogenesis in rats.
