目的:建立测定大鼠血浆中新芒果苷HPLC方法,探究盐炙对知母中新芒果苷药代动力学的影响。方法:色谱柱:Ecosil C_(18)柱(250 mm×4.6 mm,5μm);流动相:水-乙腈(15:85);流速:1 m L/min;检测波长:258 nm;柱温:30℃。结果:新芒果苷在生品和盐炙品组大鼠体内药代动力学参数分别为:AUC_(0-t)为(43.913±0.636)、(71.565±1.950)mg/L*h;AUC_(0-∞)为(44.127±0.673)、(71.805±1.926)mg/L*h;C_(max)为(4.539±0.048)、(5.805±0.119)mg/L;MRT_(0-t)为(8.124±0.246)、(8.650±0.398)h;MRT_(0-∞)为(8.290±0.321)、(8.761±0.435)h;t_(1/2)为(4.315±0.472)、(3.989±0.301)h。结论:知母盐炙后促进新芒果苷的吸收,并在一定程度上增加了生物利用度。
OBJECTIVE: To establish a HPLC method for the determination of nematanin in rat plasma and explore the effect of saltburn on the pharmacokinetics of ne mangiferin in Zhimu. Method: Column: Ecosil C 18 column (250 mm × 4.6 mm, 5 μm); mobile phase: water-acetonitrile (15:85); flow rate: 1 m L / min; detection wavelength: 30 ° C. Results: The pharmacokinetic parameters of new mangiferin in the raw and salt Sunburn products were AUC 0-t (43.913 ± 0.636) and (71.565 ± 1.950) mg / L * (4.5 ± 0.048) and (5.805 ± 0.119) mg / L, respectively; (3) the average value of MRT_ (0-t) was 0.343 ± 0.673 and (71.805 ± 1.926) mg / L * 8.124 ± 0.246, and 8.650 ± 0.398 h, respectively; MRT_ (0-∞) was (8.290 ± 0.321) and (8.761 ± 0.435) h respectively; t 1/2 was (4.315 ± 0.472) and (3.989 ± 0.301) h Conclusion: Zhimu salt after Sunburn to promote the absorption of new mangiferin, and to some extent, increased bioavailability.
