本文报道了B族链球菌(GBS)Ib型多糖-破伤风类毒素(lb-TT)结合菌苗的合成及其在兔体内的免疫原性.此外,将Ib-TT菌苗同Ia、Ⅱ和Ⅲ型多糖与TT结合的三价菌苗合成一种四价GBS结合菌苗,通过建立GBS感染的母鼠免疫一新生鼠攻击模型.确定该四价结合菌菌对新生鼠的保护功效.采用化学方法将TT与GBS荚膜型特异性多糖上唾液酸残基共价结合,制备成GBS Ib-TT结合菌苗及GBS四价结合菌苗.用Ib-TT菌苗皮下注射免疫新西兰白兔,以ELISA法测定荚膜特异性抗体.母鼠腹腔内注射四价结合菌苗,于新生鼠出生后36小时以GBS作腹腔内攻击,测定四价结合菌苗对新生鼠的保护作用.
In this paper, the synthesis of IBS-TB-TB-TB vaccine and its immunogenicity in rabbits were reported.In addition, And GB Ⅲ -activated trivalent bacterium vaccine to synthesize a tetravalent GBS-conjugated bacterin and immunize a neonatal rat challenge model by establishing GBS-infected female mice to determine the protective effect of the tetravalent conjugate bacterium on newborn mice. The GBS Ib-TT conjugate vaccine and GBS quadrivalent bacterin were prepared by covalent attachment of TT and GBS capsular polysaccharide to sialic acid residues by chemical method.The New Zealand white was immunized with Ib-TT vaccine subcutaneously The rabbits were tested for capsular specificity antibody by ELISA method.Mice were intraperitoneally injected with tetravalent conjugate bacterin and intraperitoneally challenged with GBS 36 hours after birth to determine the protective effect of tetravalent conjugate vaccine on neonatal rats.
