目的研究N-糖基化抑制对胶质瘤细胞生长状态的影响。方法以依霉素(tunicamycin)为N-糖基化抑制剂,作用于人脑胶质瘤细胞株SWO-38,运用电镜技术及琼脂糖电泳,观察N-糖基化抑制对胶质瘤细胞生长的影响。结果依霉素作用于SWO-38细胞后,透射电镜下细胞出现线粒体等细胞器增殖,核仁消失,细胞团缩。琼脂糖电泳出现DNA梯形条带。结论N-糖基化抑制可以诱导胶质瘤细胞的早期凋亡。
Objective To investigate the effect of N-glycosylation on the growth of glioma cells. Methods The human glioma cell line SWO-38 was treated with tunicamycin as an inhibitor of N-glycosylation. DNA trapezoidal bands appeared on agarose gel electrophoresis. Conclusion N-glycosylation can induce early apoptosis of glioma cells.
