目的:建立保健食品中大豆苷、大豆黄素、染料木苷、大豆素、大豆黄苷、染料木素和葛根素的高效液相色谱同时测定方法。方法:样品经70%乙醇超声提取,过滤膜后进行液相色谱测定,液体样品过滤后直接测定。色谱柱为C18(250 mm×4.6 mm,5μm)柱;流动相为甲醇和1%磷酸水溶液梯度洗脱;流速为1.0 ml/min;柱温为30℃;二级管阵列检测器检测波长为254 nm。结果:方法回收率为97.5%~105%,RSD为0.46%~3.33%,检出限分别为0.26μg/ml~0.60μg/ml。结论:该方法简便、快速、重现性好,可用于保健食品中这6种大豆异黄酮和葛根素的同时测定。
Objective: To establish a method for simultaneous determination of daidzin, daidzein, genistin, daidzein, daidzein, genistein and puerarin in health food by high performance liquid chromatography. Methods: The samples were extracted by ultrasonic with 70% ethanol. After the membrane was filtered, liquid chromatography was performed. The liquid samples were directly filtered after filtration. The column was C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was gradient elution of methanol and 1% phosphoric acid. The flow rate was 1.0 ml / min. The column temperature was 30 ℃. The detection wavelength of the diode array detector 254 nm. Results: The recoveries were 97.5% -105% with RSDs of 0.46% -3.33%. The detection limits were 0.26μg / ml ~ 0.60μg / ml respectively. Conclusion: The method is simple, rapid and reproducible. It can be used for simultaneous determination of six isoflavones and puerarin in health food.
