为时刻监控治疗肝癌时的抗癌药物剂量即血中的药物浓度 ,提高疗效 ,减少副作用 ,采用紫外 - HPLC法对血中和组织中的阿霉素浓度进行分析 .用 UV- 2 4 0紫外分光光度计 ,BECKMAN332型高效液相色谱仪 ,采用反向色谱法 .结果表明 ,阿霉素在 2 33nm、2 5 4 nm、2 90 nm出现最大吸收峰 ,在 2 4 3nm、2 80 nm处出现最小吸收峰 ,标准品在 ( 2~ 2 0 )μg/m L呈直线关系 ,阿霉素和内标色谱保留时间分别为 5 min和 7min.血浆阿霉素浓度在 ( 5 0~ 1 0 0 ) μg/m L范围内呈良好的线性关系 ,血浆阿霉素平均回收率为 96.2 % ,并得出肝、肾、肠、心肌组织阿霉素的回归直线方程
To monitor the time of treatment of liver cancer anticancer drug dose that blood concentration of the drug to improve efficacy and reduce side effects, using UV - HPLC method in the blood and tissue adriamycin concentrations were analyzed with UV-2 0 Spectrophotometer and BECKMAN332 high performance liquid chromatography with reverse phase chromatography.The results showed that the maximum absorption peak of adriamycin at 2 33nm, 2 54nm and 2 90 nm appeared at 2 43nm and 2 80nm (2 ~ 20) μg / m L, the retention times of doxorubicin and internal standard were 5 min and 7 min respectively.The plasma doxorubicin concentration was in the range of (50 ~ 1 0 0) μg / m L, the average recovery rate of doxorubicin in plasma was 96.2%, and the linear regression equation of doxorubicin in liver, kidney, intestine and myocardium was obtained
