黄芪血清药理对HDF细胞p16mRNA表达及端粒长度影响的实验研究

目的:观察黄芪血清对人胚肺二倍成纤维(HDF)细胞超氧化物歧化酶(SOD)活性、p16mRNA表达及端粒长度影响。方法:以黄芪水煎剂灌胃家兔获得含药血清,以含黄芪血清的DMEM培养基培养HDF细胞,同时以不含黄芪的家兔血清作空白对照,比色法检测HDF细胞的SOD活性、RT-PCR检测p16mRNA表达及TRF-Southernblot法检测HDF细胞端粒长度变化。结果:含黄芪血清培养基培养的HDF细胞与空白血清培养的HDF细胞比较SOD活性增加,p16mRNA表达减少,端粒酶长度增加。结论:含黄芪血清的培养基通过增加HDF细胞SOD活性,减少p16mRNA表达,抑制端粒缩短而起到抗衰老作用。 OBJECTIVE: To observe the effect of serum of Astragalus on SOD activity, p16 mRNA expression and telomere length in human embryo lung doubled fibroblast (HDF) cells. Methods: Rabbits were intragastrically administered with Astragalus decoction to obtain drug-containing serum. HDF cells were cultured in DMEM medium containing Astragalus membranaceus serum. The serum of rabbits without Astragalus membranaceus was used as a blank control. The SOD activity of HDF cells was detected by colorimetry. RT-PCR was used to detect the expression of p16 mRNA and TRF-Southern blot to detect the change of telomere length in HDF cells. RESULTS: The activity of SOD, the expression of p16 mRNA and the length of telomerase were increased in HDF cells cultured with Astragalus membranaceus and HDF cells cultured in blank serum. Conclusion: The medium containing Astragalus membranaceus could increase the activity of SOD in HDF cells, reduce the expression of p16 mRNA, and inhibit the shortening of telomeres to play an anti-aging effect.
