目的探讨补肾中药对双侧卵巢摘除引起的骨质疏松大鼠m GluR5表达的影响。方法取3月龄SPF级雌性SD大鼠40只,随机分为假手术组(Sham group)、模型组(Model group)、中药组(TCM group)、雌激素药物治疗组(E group),每组10只。对sham组大鼠施行假手术,其余4组行双侧卵巢摘除术。术后30d进行相应的给药实验,TCM组按10g/kg生药剂量灌胃,E组按0.2mg/kg剂量灌胃,Sham及Model组灌服等量的生理盐水。12周后测定各组大鼠血清中I型胶原羧基端肽(β-Crosslaps)、总I型胶原氨基端延长肽(P1NP)、Ca和P的含量;以双能X线骨密度测定仪测定大鼠的骨密度;Western blot方法检测股骨骨骺m GluR5蛋白表达。结果 Model组BMD为(0.137±0.013)g/cm2,血清Ca/P比值为(1.65±0.37)、P1NP浓度(4.388±0.400)ng/m L、Glu表达(0.424±0.061)与假手术组比较显著降低(P<0.05),β-Crosslaps浓度(0.745±0.075)ng/m L、m GluR5表达(0.855±0.116)较假手术组高(P<0.01);TCM组BMD为(0.156±0.006)g/cm2,血清Ca/P比值为(2.59±0.55)、P1NP浓度为(5.749±0.611)ng/m L、Glu表达(0.702±0.080)与模型组比较显著增高(P<0.01),β-Crosslaps浓度(0.648±0.069)ng/m L、m GluR5表达(0.553±0.070)显著降低(P<0.01);E组组BMD(0.146±0.006)g/cm2、Glu表达(0.547±0.075)低于TCM组(P<0.05),而其β-Crosslaps浓度为(0.649±0.052)ng/m L、P1NP浓度为(5.516±0.351)ng/m L、Ca/P比值为(2.70±0.55)则与TCM组没有差异(P>0.05)。结论补肾中药能有效防止去卵巢模型大鼠骨质疏松的形成,可能通过调节骨内谷氨酸进一步介导骨重建而发挥作用。
Objective To investigate the effect of Bushen Chinese medicine on the expression of m GluR5 in rats with osteoporosis induced by bilateral ovariectomy. Methods Forty SPF female SD rats of 3 months old were randomly divided into Sham group, Model group, TCM group and E group. Group of 10. Conclusion Bushen Chinese medicine can effectively prevent the formation of osteoporosis in ovariectomized rats may play a role by regulating the bone glutamate further mediated bone remodeling.
