目的:观察温胆汤加味方对糖尿病性脑梗死(DACI)大鼠脑匀浆一氧化氮(NO)、一氧化氮合酶(NOS)的影响。方法:实验共设六组:假手术组、模型组、温胆汤加味方大、中、小剂量组及对照组(血塞通)。采用链脲佐菌素制造糖尿病模型,血糖≥11.1mmol/L者4周后,建立大脑中动脉阻塞(MCAo)模型。灌药2周后,检测各组脑匀浆一氧化氮(NO)含量及一氧化氮合酶(NOS)活性。结果:应用温胆汤加味方后大鼠脑匀浆中一氧化氮(NO)含量、一氧化氮合酶(NOS)活性有不同程度的下降(P均<0.05),以大、中剂量组为优。结论:大、中剂量温胆汤加味方能够抑制内皮细胞、星形胶质细胞、脑巨噬细胞表达一氧化氮合酶(NOS),减少一氧化氮(NO)释放,减轻内皮细胞损伤与氧化应激。
OBJECTIVE: To observe the effects of Wendan Tang Jiawei Recipe on nitric oxide (NO) and nitric oxide synthase (NOS) in brain homogenate of rats with diabetic cerebral infarction (DACI). Methods: A total of six groups were set up in the experiment: sham operation group, model group, Wendan Decoction plus large, medium and small dose group and control group ( Xuesaitong). A diabetic model was made using streptozotocin. After 4 weeks of blood glucose ≥ 11.1 mmol/L, a middle cerebral artery occlusion (MCAo) model was established. After 2 weeks of dosing, the content of nitric oxide (NO) and the activity of nitric oxide synthase (NOS) were measured in brain homogenates of each group. Results: The content of nitric oxide (NO) and the activity of nitric oxide synthase (NOS) in the brain homogenate were decreased in different degrees after application of Wendan Decoction (P<0.05). The large and medium dose groups were excellent. Conclusion: Large and medium doses of Wendan Decoction can inhibit the expression of nitric oxide synthase (NOS), reduce nitric oxide (NO) release, and reduce endothelial cell injury and oxidation in endothelial cells, astrocytes, and macrophages.
