目的探讨旱莲草水提取物对高同型半胱氨酸血症(Hhcy)大鼠血脂及血管内膜的影响。方法通过喂饲高L-蛋氨酸饲料,复制HHcy大鼠模型。50只SD大鼠随机分为5组,正常组、模型组、洛伐他汀组(2 mg·kg-1)及旱莲草低、高2个剂量组(5,10 g·kg-1),每日灌胃给药。8周后,生化法测定血清同型半胱氨酸(Hcy)、总胆固醇(TC)、三酰甘油(TG)水平,酶联免疫吸附法检测一氧化氮(NO)、内皮素(ET)及血栓素B2(TXB2)水平,胸主动脉切片HE染色在光镜下观察血管形态学改变。结果与模型组相比,高剂量旱莲草组血清NO水平明显上升,其余各项指标均显著降低,胸主动脉血管形态基本正常。结论旱莲草水提取物能够有效降低Hhcy大鼠血清Hcy水平,具有调节血脂、保护血管内膜的作用。
Objective To investigate the effects of Eclipta prostrata extract on blood lipids and vascular intima in hyperhomocysteinemia rats. Methods HHcy rat model was reproduced by feeding high L-methionine diet. Fifty Sprague-Dawley rats were randomly divided into 5 groups: normal group, model group, Lovastatin group (2 mg · kg -1) and Eclipta prostrata (2 × 10 ^ g · kg -1) , Daily gavage. After 8 weeks, serum homocysteine (Hcy), total cholesterol (TC) and triglyceride (TG) levels were measured by biochemical method. Nitric oxide (NO), endothelin (ET) and enzyme linked immunosorbent assay Thromboxane B2 (TXB2) levels and thoracic aorta sections were observed under light microscope for morphological changes of blood vessels. Results Compared with the model group, the serum NO level of high dose Eclipta prostrata group significantly increased, the rest of the indicators were significantly reduced, the morphology of thoracic aorta was normal. Conclusion Eclipta prostrata extract can effectively reduce the level of serum Hcy in Hhcy rats, and has the effect of regulating blood lipids and protecting the intima of blood vessels.
