目的通过实时测定单态氧(singlet oxygen,1 O2)评估635nm和(或)670nm激光照射下盐酸氨基乙酰丙酸(5-aminolevulinic acid,5-ALA)介导的光动力学治疗(photodynamic therapy,PDT)抗肿瘤效果。方法以5-ALA为光敏剂,在单波长(635nm或670nm)及联合波长(635nm+670nm)下进行激光照射,利用新型高灵敏的检测系统测定PpIX溶液、肿瘤细胞(9L神经胶质肉瘤细胞和HeLa细胞)及HeLa负瘤裸鼠中的1 O2强度,H-E染色法检测肿瘤组织坏死深度。结果接受单一的635nm或670nm波长激光照射时,PpIX溶液1 O2强度在两者中无明显区别。635nm与670nm激光联合照射下,9L细胞的1 O2强度高于HeLa细胞;在两种细胞中670nm激光诱发1 O2强度均远远低于635nm激光。在HeLa细胞裸鼠皮下移植瘤中,两种波长激光联合照射产生一定的肿瘤坏死深度,但低于635nm单波长照射。结论在5-ALA介导的PDT中,670nm波长激光显示了一定的PDT疗效。
Objective To evaluate the photodynamic therapy of 5-aminolevulinic acid (5-ALA) mediated by 635nm and 670nm laser irradiation by real-time measurement of singlet oxygen (1 O2) PDT) anti-tumor effect. Methods 5-ALA was used as a photosensitizer and laser irradiation was performed at a single wavelength (635nm or 670nm) and a combined wavelength (635nm + 670nm). PpIX solution, tumor cells (9L glioma sarcoma cells And HeLa cells) and HeLa negative nude mice in 1 O 2 intensity, HE staining of tumor necrosis depth. Results When exposed to a single laser at 635 nm or 670 nm, there was no significant difference in the intensity of 1 P 2 in PpIX solution. The intensity of 1 O2 in 9L cells was higher than that in HeLa cells when exposed to 635nm and 670nm laser. The intensity of 1O2 induced by 670nm laser in both cells was much lower than that of 635nm laser. In HeLa nude mice subcutaneously transplanted tumor, the combination of two wavelength laser irradiation produced a certain depth of tumor necrosis, but lower than 635nm single wavelength irradiation. Conclusions In the 5-ALA-mediated PDT, 670 nm wavelength laser showed some PDT efficacy.
