目的从T细胞中获得更多含PH结构域编码序列的新基因。方法 采用低严谨核酸杂交技术,根据T细胞中已知的PH结构域的分子设计探针,对人T细胞淋巴瘤cDNA文库进行筛选。结果 在筛选过程中得到了4个新基因,各cDNA长度依次为:1733bp、2308bp、2130bp和1647bp,分别编码含229、376、211 和 143个氨基酸残基的多肽。经生物信息学分析,在基因数据库中未发现与此4种cDNA序列类似的基因。该4个基因均已被GenBank收录,登录号分别为:AF334588AF134589、AF334590和 AF334792。结论 这些新基因的获得及进一步研究,可能会为深人探讨T细胞激活机制提供新的线索。
OBJECTIVE: To obtain more new genes containing the coding sequence of PH domain from T cells. Methods A low-stringency nucleic acid hybridization technique was used to screen the human T-cell lymphoma cDNA library based on the molecular design probes of known PH domains in T cells. Results Four novel genes were obtained. The length of each cDNA was 1733bp, 2308bp, 2130bp and 1647bp, encoding 229, 376, 211 and 143 amino acid residues respectively. After bioinformatics analysis, no gene similar to the four cDNA sequences was found in the gene database. The four genes have been deposited in GenBank accession number: AF334588AF134589, AF334590 and AF334792. Conclusion The gain and further study of these new genes may provide new clues for exploring the T cell activation mechanism in depth.
