目的从小刺猴头菌子实体中提出水溶性粗多糖(HP),从粗多糖中分离出均一多糖并研究其单糖组成。方法DEAE-SepharoseCL-6B及SephadexG-100柱色谱进一步纯化得HPⅠ,柱色谱,醋酸纤维素薄膜电泳等方法检验纯度,GC和PC测其单糖组成。结果HPⅠ为均一多糖,相对分子质量约为65000,单糖组成为:Fuc,Glc,Gal,摩尔比依次为0.423:1:2.110。结论HPⅠ为中性杂多糖,首次从小刺猴头菌子实体中提取获得。
OBJECTIVE To extract the water-soluble polysaccharide (HP) from the fruit body of Hericium erinaceus, and to isolate the uniform polysaccharide from the crude polysaccharide and study its monosaccharide composition. Methods Purified by DEAE-Sepharose CL-6B and Sephadex G-100 column chromatography. HPI, column chromatography, and cellulose acetate membrane electrophoresis were used to test the purity. GC and PC were used to determine the monosaccharide composition. Results The HPI was a homogeneous polysaccharide with a relative molecular weight of about 65000. The monosaccharide composition was: Fuc, Glc, Gal, and the molar ratio was 0.423:1:2.110 in that order. Conclusion HPI is a neutral heteropolysaccharide and was first extracted from the fruit body of Hericium erinaceus.
