以红丰和宝交早生品种的组培苗为试材,初步建立了叶圆盘和叶柄片段植株再生体系。红丰的芽苗再生能力高于宝交早生,叶圆盘的再生频率高于叶柄片段,叶圆盘下表面(背面)接触培养基比上表面(正面)更有利于再生。AgNO3有利于芽苗再生,在接种叶圆盘前加入主要刺激芽苗的直接再生,而在愈伤组织形成后加入则主要提高再生频率。再生芽苗在三角瓶中容易生根,移苗入土后生长正常。
Taking the tissue culture seedlings of Hongfeng and Baojiao-Born early breeds as test materials, the plant regeneration system of leaf disk and petiole fragments was preliminarily established. The regenerative capacity of Hongfeng seedlings was higher than that of Baojiao and Born ones, and the regeneration frequency of leaf discs was higher than that of petiole segments. The lower surface of disc leaf (back surface) was more conducive to regeneration than the upper surface (front surface). AgNO3 is beneficial to the regeneration of shoots, and the direct regeneration before the inoculation of leaf disks is mainly to stimulate sprouts, while the addition of callus after the formation of callus mainly improves the regeneration frequency. Bud sprouts in the triangle regeneration of root easily rooted, Miao Miao soil growth after normal.
