目的 :探讨中药复方益气解毒颗粒防治鼻咽癌的作用机制。方法 :用二亚硝基哌嗪 (DNP)诱导大鼠鼻咽癌动物模型 ,观察益气解毒颗粒对大鼠鼻咽癌变中病理形态改变以及端粒酶和端粒酶RNA表达的影响。定期分批处死大鼠 ,取鼻咽组织作病理学检测 ,用PCR -ELISA和NestedRT -PCR法检测大鼠鼻咽组织端粒酶的活性和端粒酶RNA的表达。结果 :灌服益气解毒颗粒大鼠鼻咽癌发癌率 0 (0 /5 ) ,明显低于盐水对照组 80 % (4/5 ) (包括原位癌和浸润癌 ) ;其端粒酶活性为 0 2 4± 0 11,也明显低于对照组 0 93± 0 2 3(P <0 .0 5 ) ;维甲酸组发病率为 0 (0 /5 ) ,端粒酶活性为 0 2 7± 0 13;各组端粒酶RNA均为阳性。维甲酸组大鼠于 2 15~ 30 8d间逐步死亡。结论 :益气解毒颗粒能阻断DNP诱导大鼠鼻咽癌变 ,且抑制鼻咽上皮癌变细胞端粒酶的激活
Objective: To investigate the mechanism of traditional Chinese medicine Yiqi Jiedu Granule in preventing and treating nasopharyngeal carcinoma. Methods: The rat model of nasopharyngeal carcinoma was induced by dinitrosopiperazine (DNP). The effect of Yiqi Jiedu Granule on the pathological changes of nasopharyngeal carcinoma and the expression of telomerase and telomerase RNA were observed. The rats were sacrificed in batches on a regular basis and the nasopharyngeal tissues were taken for pathological examination. The activity of telomerase and the expression of telomerase RNA in nasopharyngeal tissues of rats were detected by PCR-ELISA and NestedRT-PCR. Results: The rate of carcinogenesis of nasopharyngeal carcinoma was 0 (0/5), significantly lower than that of saline control group (80%) (4/5) (including carcinoma in situ and invasive carcinoma); telomerase The activity was 0 2 4 ± 0 11, which was also significantly lower than that of the control group 0 93 ± 0 2 3 (P 0 05). The incidence of retinoic acid group 0 (0/5) and telomerase activity 0 2 7 ± 0 13; telomerase RNA in each group was positive. Rats in the retinoic acid group died gradually from 2 15 to 30 8d. Conclusion: Yiqi Jiedu Granule can block the DNP-induced nasopharyngeal carcinogenesis and inhibit the activation of telomerase in the cell of nasopharyngeal epithelial carcinoma
