目的研究CBA/N、nude和SCID小鼠人肝癌模型的特点,凋亡相关基因的表达及其相关关系。方法用人肝癌细胞系HHCC9724P接种于动物肝脏,于各时间点观察肿瘤体积的生长情况。用免疫组织ABC法对其bcl2,bax和VEGF的表达及其与肿瘤组织体积相关性进行研究分析。结果3种动物肝内种植肝癌细胞后均可成活,bcl2,bax和VEGF的表达明显增强(P<0.01)。肝癌细胞种植3周后肿瘤体积和基因表达达最高水平。其中SCID小鼠尤为突出(P<0.01)。3种小鼠肝内肿瘤体积发展与3种基因表达改变相关性分析结果证明具有明显的正相关关系。结论SCID小鼠的人肝癌细胞种植成功率和凋亡基因表达最为显著。肝内肿瘤的生长与凋亡基因表达具有明显的正相关关系。
Objective To investigate the characteristics of human hepatocarcinoma model of CBA / N, nude and SCID mice and the expression of apoptosis related genes. Methods Human liver cancer cell line HHCC9724P was inoculated into the liver of animals and the growth of tumor volume was observed at each time point. Immunohistochemical ABC method for its bcl2, bax and VEGF expression and tumor tissue volume correlation analysis. Results All the three kinds of animals could survive after intrahepatic implantation of hepatoma cells, and the expressions of bcl2, bax and VEGF were significantly increased (P <0.01). Tumor volume and gene expression reached the highest level after three weeks of planting of hepatoma cells. SCID mice were particularly prominent (P <0.01). The results of the correlation analysis between the three kinds of mice intrahepatic tumor volume development and the changes of three kinds of gene expression showed a significant positive correlation. Conclusion The success rate and the expression of apoptotic genes in SCID mice are the most significant. The intrahepatic tumor growth and apoptosis gene expression has a significant positive correlation.
