针对单株DNA、多株叶片混合DNA和单株DNA混合的DNA取样方法,利用SSR技术对蒙A群和C群1进行遗传变异分析,探讨建立玉米群体遗传多样性分析的技术体系。采用均匀分布在玉米10条染色体上的34对SSR引物对不同处理的DNA样本进行扩增,通过聚丙烯酰胺凝胶电泳检测,对60个单株进行多态性信息量与遗传相似系数分析,比较不同处理的DNA样本扩增的等位基因数目。结果表明,2个供试群体均具有较丰富的遗传变异,建立的技术体系可用于群体的遗传多样性研究,采用12株叶片混合、5次重复提取的DNA样本可以代替相同数目的单株DNA混合的DNA样本,为最优取样方法。
Aiming at DNA sampling method of mixing single plant DNA and multiple leaves mixed DNA with single plant DNA, SSR technique was used to analyze the genetic variation of Meng A group and C group 1, and to establish the technical system of genetic diversity analysis of maize population. 34 pairs of SSR primers evenly distributed on 10 chromosomes of maize were used to amplify the different treated DNA samples. Polyacrylamide gel electrophoresis was used to test the polymorphism information content and genetic similarity coefficient of 60 individuals. Compare the number of alleles amplified by different treated DNA samples. The results showed that both of the two test populations had rich genetic variation. The established technology system could be used to study the genetic diversity of the population. DNA samples mixed with 12 leaves and 5 replicates could replace the same number of single plant DNA Mixed DNA samples, the best sampling method.
