p70核糖体蛋白S6激酶(p70 ribosomal protein S6 kinase,p70S6K)是一种多功能的丝氨酸/苏氨酸蛋白激酶,是AGC家族的重要成员,在m TOR信号通路中起着非常重要的作用。p70S6K与蛋白质的合成、m RNA的加工、葡萄糖的体内平衡以及细胞的生长和凋亡等进程密切相关。研究表明,肿瘤的发生与p70S6K的过度激活有关,因此,p70S6K已成为抗癌药物研发的重要潜在靶点之一。本文对p70S6K的生物学功能、作用机制及相关p70S6K抑制剂结构和研究思路做一综述。
p70 ribosomal protein S6 kinase (p70S6K) is a multifunctional serine / threonine protein kinase and is an important member of the AGC family. It plays a very important role in the mTOR signaling pathway. p70S6K and protein synthesis, m RNA processing, glucose homeostasis and cell growth and apoptosis processes are closely related. Studies have shown that the occurrence of cancer and p70S6K over-activation, therefore, p70S6K has become one of the important potential targets for the development of anti-cancer drugs. This review summarizes the biological functions of p70S6K, its mechanism of action and related p70S6K inhibitor structure and research ideas.
