目的:观察加味消渴康对糖尿病肾病大鼠肾组织MMP-9和TIMP-1表达的影响。方法:采用单侧肾切除加链脲佐菌素诱导的DN大鼠模型。实验随机分为正常组、模型组、氯沙坦组以及加味消渴康大、中、小三个剂量组并给予药物干预。各组大鼠于第12周末处死取出肾脏,运用免疫组化法观察肾组织MMP-9和TIMP-1的表达。结果:与正常组相比,模型组MMP-9表达减弱;TIMP-1表达增强。氯沙坦组、加味消渴康三个剂量组MMP-9表达较模型组增强,积分光密度值升高;TIMP-1表达减弱,积分光密度值降低,差异有统计学意义(P<0.01)。结论:加味消渴康对DN大鼠MMP-9和TIMP-1表达有一定的调节作用。
Objective: To observe the effect of Jiawei Xiaokekang on the expression of MMP-9 and TIMP-1 in the kidney of diabetic nephropathy rats. Methods: Unilateral nephrectomy with streptozotocin-induced DN rat model. The experiment was randomly divided into normal group, model group, losartan group and Jiawei Xiaoke Kang large, medium and small dose groups and given drug intervention. The rats were sacrificed at the end of the twelfth week and the kidneys were removed. The expression of MMP-9 and TIMP-1 in renal tissues was observed by immunohistochemistry. Results: Compared with the normal group, the expression of MMP-9 in the model group decreased and the expression of TIMP-1 increased. Compared with the model group, the expression of MMP-9 in losartan group and Jiawei Xiaokekang group were higher than that in model group, and the integral optical density value was increased; the expression of TIMP-1 was weakened and the integral optical density value was decreased, the difference was statistically significant (P <0.01 ). Conclusion: Jiawei Xiaoke Kang can regulate the expression of MMP-9 and TIMP-1 in DN rats.
