目的:探讨三苯氧胺(Tamoxifen,TAM)对人舌鳞癌细胞系Tca8113的增殖抑制作用。方法:不同浓度TAM处理体外培养的人舌鳞癌细胞Tca8113,MTT法检测细胞增殖,倒置显微镜下观察细胞形态学变化。结果:在浓度为1、2、5、10μmol/L时,TAM呈现出剂量依赖性抑制人舌鳞癌细胞Tca8113的增殖作用。通过对三个不同浓度(2μmol/L、5μmol/L、10μmol/L)TAM实验组的抑制率进行比较,差异有统计学意义(P<0.05),细胞抑制率与药物浓度呈正相关。细胞超微结构显示,药物浓度与细胞结构呈剂量依赖关系。结论:TAM可显著抑制人舌鳞癌细胞Tca8113的增殖,其作用呈剂量依赖关系。
Objective: To investigate the inhibitory effect of Tamoxifen (TAM) on human tongue squamous cell carcinoma cell line Tca8113. Methods: Human tongue squamous carcinoma cell line Tca8113 cultured in vitro was treated with different concentrations of TAM. Cell proliferation was measured by MTT assay. Morphological changes were observed under inverted microscope. Results: At concentrations of 1, 2, 5 and 10 μmol / L, TAM showed a dose-dependent inhibition of Tca8113 proliferation in human tongue squamous cell carcinoma. The inhibitory rates of TAM in three different concentrations (2μmol / L, 5μmol / L, 10μmol / L) were significantly different (P <0.05). The inhibition rate was positively correlated with drug concentration. Cell ultrastructure showed that drug concentration and cell structure in a dose-dependent manner. Conclusion: TAM can significantly inhibit the proliferation of human tongue squamous cell carcinoma Tca8113 cells in a dose-dependent manner.
