目的:定量研究嘌呤霉素肾病大鼠模型从建立到恢复过程中,足突的形态变化规律。方法:用透射电镜摄片,图像分析仪分析并应用形态计量学的方法,研究了33例大鼠(嘌呤霉素注射实验组及生理盐水注射对照组)不同时间点的足突形态变化。结果:注射嘌呤霉素后,足突表现出由变宽到融合又逐渐恢复的形态变化,形态计量学的结果进一步表明,各时间点实验组足突宽度,体积密度,比表面与对照组相比有高度显著性差异(P<0.01),表面积密度在2--15天与对照组差异显著(P<0.05)。且实验组内相邻的不同时间点间比较, 足突宽度、体积密度、表面积密度、 比表面差异均具有高度统计学意义(P<0.01)。结论:形态计量学对嘌呤霉素肾病足突形态研究,定量表现出足突由变宽到融合以后逐渐恢复的动态过程,为今后对功能与分子水平的研究提供早期准确的实验形态依据。
OBJECTIVE: To quantitatively study the morphological changes of foot process in puromycin nephropathy rat model from establishment to recovery. Methods: The morphological changes of the foot process in 33 rats (puromycin injection group and saline injection control group) were studied by transmission electron microscope (TEM) and image analyzer. RESULTS: After injection of puromycin, the process of foot process showed morphological change from broadening to fusion and gradual recovery. The results of morphometry further showed that the width of the foot process, the bulk density and the specific surface area (P <0.01). The surface area density was significantly different from that in the control group (P <0.05) in the range of 2-15 days. Compared with the adjacent groups at different time points in the experimental group, foot width, bulk density, surface area density and specific surface area were highly statistically significant (P <0.01). CONCLUSIONS: Morphometric studies on the morphology of foot processes in puromycin nephropathy quantitatively show the dynamic process of foot process width broadening to fusion. It provides an accurate and early experimental basis for the study of function and molecular level in the future.
