用膜片钳单通道技术 ,观察在细胞膜内侧加入甲基莲心碱 (Nef)后 K+通道特性的改变 ,以研究Nef对豚鼠心室肌细胞及猪冠状动脉平滑肌细胞K+通道的作用 .结果 :Nef可使心肌细胞及冠脉平滑肌细胞上 K+通道开放概率 (PO)降低 . 1 0 ,2 0μmol· L-1Nef分别使心肌细胞 K+通道降低 (50± 32 ) %和 (96± 5) % ,30 ,50 μmol· L-1的 Nef分别使猪冠脉平滑肌细胞上 K+通道 PO 降低 (51±2 1 ) %和 (71± 1 4) % .结果表明 ,Nef可从通道内口阻断心肌及平滑肌细胞上 K+通道 ,但对前者的作用更强
To observe the effect of Nef on the K + channels of guinea pig ventricular myocytes and porcine coronary artery smooth muscle cells by patch clamp single-channel technique to observe the changes of K + channels after adding Nef into the cell membrane.Results: The K + channel opening probability (PO) of myocardial cells and coronary artery smooth muscle cells was decreased.On 10, 20μmol · L-1Nef decreased K + channels in cardiomyocytes by (50 ± 32)% and (96 ± 5)%, Nef at 50 μmol · L-1 decreased (51 ± 21)% and (71 ± 14)% PO of K + channels in porcine coronary artery smooth muscle cells, respectively.
