目的:顺铂作为传统的化疗药物被广泛应用于临床,但其较强的副作用限制了顺铂的临床应用。为了提高肿瘤细胞对顺铂的敏感,我们选择2-脱氧-D-葡萄糖(2-DG)进行细胞实验。方法:利用免疫荧光结合免疫印迹技术检测2-脱氧-D-葡萄糖(2-DG)体外抗卵巢癌SKOV3细胞的抗肿瘤作用。结果:2-DG可增强顺铂对卵巢癌SKOV3细胞的抗肿瘤作用,包括抑制SKOV3细胞的增殖、诱导凋亡小体的形成和促进细胞的凋亡。免疫荧光结合免疫印迹技术检测显示,2-DG增强了顺铂诱导的SKOV3细胞内质网应激相关蛋白--葡萄糖调节蛋白78(GRP78)的表达;2-DG还上调顺铂诱导的SKOV3细胞内质网应激相关蛋白--凋亡蛋白153/C/EBP同源蛋白(CHOP)的表达。结论:2-DG通过增强顺铂诱导的内质网应激增加SKOV3细胞对顺铂的敏感性。
Objective: Cisplatin as a traditional chemotherapy drug is widely used in clinical practice, but its strong side effects limit the clinical application of cisplatin. To increase the sensitivity of tumor cells to cisplatin, we selected 2-deoxy-D-glucose (2-DG) for cell experiments. Methods: The anti-tumor effects of 2-deoxy-D-glucose (2-DG) against ovarian cancer SKOV3 cells in vitro were detected by immunofluorescence combined with immunoblotting. Results: 2-DG can enhance the antitumor effect of cisplatin on ovarian cancer SKOV3 cells, including inhibiting the proliferation of SKOV3 cells, inducing the formation of apoptotic bodies and promoting cell apoptosis. Immunofluorescence combined with Western blotting showed that 2-DG enhanced cisplatin-induced expression of endoplasmic reticulum stress-related protein Glucose Regulatory Protein 78 (GRP78) in SKOV3 cells; 2-DG also up-regulated cisplatin-induced SKOV3 cells Endoplasmic reticulum stress related protein - the expression of apoptosis protein 153 / C / EBP homologous protein (CHOP). CONCLUSION: 2-DG increases the sensitivity of SKOV3 cells to cisplatin by increasing cisplatin-induced endoplasmic reticulum stress.
