目的研究三邻甲苯基磷酸酯(TOCP)对人成神经瘤SH-SY5Y细胞蛋白激酶C(PKC)活性和转位的影响。方法以TOCP(0、0.25、0.5、1.0 mmol/L)对人成神经瘤细胞系SH-SY5Y细胞进行染毒,染毒时间为12、24和48 h,用PKC活性检测试剂盒检测PKC的活性,间接免疫荧光术检测PKC在细胞内的定位,荧光倒置显微镜观察结果。结果 1.0 mmol/L TOCP作用SH-SY5Y细胞24 h,PKC的活性最强,并能导致PKC由胞质向胞膜转位。结论 TOCP作用SH-SY5Y细胞后,能激活PKC,增加膜转位。
Objective To investigate the effect of tri-o-tolyl phosphate (TOCP) on protein kinase C (PKC) activity and translocation in human neuroblastoma SH-SY5Y cells. Methods Human neuroblastoma cell line SH-SY5Y cells were exposed to TOCP (0,0.25,0.5,1.0 mmol / L) for 12, 24, and 48 hours. PKC activity was measured using PKC activity assay kit Activity and indirect immunofluorescence were used to detect the localization of PKC in cells. Fluorescence inverted microscope was used to observe the results. Results After treated with 1.0 mmol / L TOCP for 24 h, the activity of PKC was the strongest and led to the translocation of PKC from cytoplasm to membrane. Conclusion TOCP can activate PKC and increase membrane translocation in SH-SY5Y cells.
