目的探讨三磷酸腺苷(ATP)结合转运蛋白G超家族成员2(ABCG2)在白血病细胞膜上的表达规律及其意义。方法应用CD45/SSC参数设门多色流式细胞术分析61例白血病患者白血病细胞膜ABCG2及其他白血病细胞相关抗原的表达情况。结果61例急慢性白血病患者的白血病细胞ABCG2表达阳性率为36.07%。ABCG2表达阳性率,急性白血病(AL)患者眼34.48%(20/58)演与慢性白血病(CL)患者眼66.67%(2/3)演鸦儿童患者眼26.09%(6/23)演与成人患者眼42.11%(16/38)演差别均无显著性意义(均P>0.05);急性髓细胞性白血病(AML)患者眼46.67%(14/30)演高于急性淋巴细胞性白血病(ALL)患者眼16.67%(4/24)(P<0.05)演;AML患儿眼55.56%(5/9)演也高于ALL患儿眼7.14%(1/14)(P<0.05)演。ABCG2与另一耐药蛋白P糖蛋白(P-gp)及其他细胞表面抗原的表达均无关(r=0.152、-0.075、-0.063、0.026、-0.149,均P>0.05),与髓系标志CD13的表达有关(r=0.307熏P<0.05)。结论ABCG2在AML患儿中的表达较高,其高表达可能导致AML患儿临床耐药。ABCG2和P-gp可能参与了不同的白血病耐药机制。
Objective To investigate the expression of ABCG2 in leukemia cell membrane and its significance. Methods The expression of ABCG2 and other leukemia-associated antigens in leukemia cell membrane of 61 patients with leukemia were analyzed by CD45 / SSC with multi-color flow cytometry. Results The positive rate of ABCG2 in leukemia cells in 61 patients with acute and chronic leukemia was 36.07%. The positive rate of ABCG2 expression was 34.48% (20/58) in acute leukemia (AL) patients and 66.67% (2/3) in chronic leukemia patients eyes. 26.09% (6/23) There was no significant difference in 42.11% (16/38) patients eyes (all P> 0.05). Acute myelogenous leukemia (AML) 46.67% (14/30) patients had higher AML than acute lymphoblastic leukemia ) In patients with AML (16.67%, 4/24) (P <0.05); 55.56% (5/9) in AML patients were also significantly higher than 7.14% (1/14) in patients with ALL (P <0.05). ABCG2 had no correlation with the expression of P-gp and other cell surface antigens (r = 0.152, -0.075, -0.063,0.026, -0.149, all P> 0.05) CD13 expression (r = 0.307 smoked P <0.05). Conclusion ABCG2 is highly expressed in children with AML, and its high expression may lead to clinical drug resistance in children with AML. ABCG2 and P-gp may participate in different mechanisms of leukemia resistance.
