目的研究单核细胞在生殖器疱疹发病机理中的作用。方法采用ELISA法,检测27例生殖疱疹患者外周血单核细胞经内毒素(LPS)诱生后培养上清中TNF-α、IL-6水平;采用APAAP 法,检测生殖器疱疹患者外周血单核细胞HLA-Ⅱ类抗原表达水平。结果生殖器疱疹患者外周血单核细胞分泌TNF-α、IL-6水平分别为3.13±0.44ng/ml和3.32±1.06ng/ml,较正常对照组[分别为(4.68±0.54ng/m1)和(6.46±1.94ng/m1)]明显降低,差异有显著性意义(P<0.05); 生殖器疱疹患者外周血单核细胞HLA-DR、HLA-DQ表达水平分别为67.48%±1.51%和29.54 %±1.15%,较正常对照组[分别为(81.03%±1.32%)和(37.63%±1.79%)]明显降低,差异有显著性意义(P<0.01)。结论单核细胞功能降低与生殖器疱疹发病有关,提高单核细胞功能, 对生殖器疱疹的防治,减少复发率有一定意义。
Objective To study the role of monocytes in the pathogenesis of genital herpes. Methods The levels of TNF-α and IL-6 in the culture supernatant of peripheral blood mononuclear cells of 27 patients with genital herpes after induced by endotoxin (LPS) were detected by ELISA. APAAP was used to detect the levels of peripheral blood mononuclear HLA-Ⅱ antigen expression level of cells. Results The levels of TNF-α and IL-6 secreted by peripheral blood mononuclear cells in genital herpes patients were 3.13 ± 0.44ng / ml and 3.32 ± 1.06ng / ml respectively, which were 4.68 ± 0.54ng / ml and (6.46 ± 1.94ng / m1)], the difference was significant (P <0.05). The levels of HLA-DR and HLA-DQ in peripheral blood mononuclear cells of patients with genital herpes were 67.48% ± 1.51% and 29.54% ± 1.15%, which was significantly lower than that of the control group [(81.03% ± 1.32%) and (37.63% ± 1.79%) respectively, with significant difference (P <0.01). Conclusions The decrease of monocyte function is related to the pathogenesis of genital herpes and the function of monocyte is improved. It is of certain significance to prevent genital herpes and reduce the recurrence rate.
