基因表达的系统分析技术 (SAGE)是一种全面分析基因表达模式的实验方法。该方法的原理是基于分离出每一m RNA所特有的长 9bp~ 14 bp的标签并将其串联起来进行克隆和测序。该方法不仅可以给出我们一个全面的特定细胞或组织的基因表达谱 ,而且还可以通过对两组处于不同环境中的同一类型细胞的基因表达谱进行比较 ,帮助我们发现那些由于环境的变化而表达发生改变的基因。本文拟就这一高通量方法的原理、应用、不足及其改进等方面的研究情况作以综述
Systemic Analysis of Gene Expression (SAGE) is an experimental method for comprehensive analysis of gene expression patterns. The principle of this method is based on the separation of the specific 9 bp to 14 bp tags unique to each m RNA and tandemly ligating them for cloning and sequencing. This method not only gives us a comprehensive gene expression profile of specific cells or tissues, but also compares gene expression profiles of two groups of cells of the same type in different environments to help us find out which of the following are due to environmental changes Genes that express changes. This article is intended to summarize the research on the principle, application, deficiency and improvement of this high-throughput method
