目的测定殃芪巴布剂中野黄芩苷、党参炔苷和延胡索乙素离体大鼠透皮率测定研究。方法采用改良的France扩散池,以离体大鼠皮肤为屏障,采用HPLC分别测定殃芪巴布剂中不同时间野黄芩苷、党参炔苷和延胡索乙素的透皮累积量。结果对殃芪巴布剂线性关系考察中,野黄芩苷回归方程为Y=3.29×10~5X-3.11×10~4(r=0.9998),结果表明野黄芩苷进样量在0.08204~0.8204μg浓度范围内线性关系良好;党参炔苷回归方程为Y=3.57×10~6X-8.54×10~4(r=0.9995),结果表明党参炔苷进样量在0.1219~1.219μg范围内线性关系良好;延胡索乙素回归方程为Y=6.54×10~6X+1.55×10~4(r=0.9996)。结果表明延胡索乙素进样量在0.02001~0.2001μg范围内线性关系良好,HPLC测得殃芪巴布剂中的野黄芩苷、党参炔苷、延胡索乙素透过离体大鼠皮肤12h透皮率分别为20.06%、19.23%、28.81%。结论殃芪巴布剂的大鼠体外经皮渗透行为是一个被动扩散行为,随着药物浓度的增加,透过量也增加。
Aim To determine the transdermal rate of Scutellaria baicalein, Codonopynin and Tetrahydropalmatine in rats. Methods A modified France diffusion cell was used to screen the skin of isolated rat skin. HPLC was used to determine the transdermal accumulation of Scutellarin, Codonopin and Tetrahydropalmatine in different time periods. Results The linear regression equation of baicalin was: Y = 3.29 × 10 ~ 5X-3.11 × 10 ~ 4 (r = 0.9998). The results showed that the baicalin of wild baicalin was 0.08204 ~ 0.8204μg The linear regression equation of Codonopin was Y = 3.57 × 10 ~ 6X-8.54 × 10 ~ 4 (r = 0.9995). The results showed that there was a good linear relationship between the dose of Ciprofloxacin and 0.1219 ~ 1.219μg The regression equation of tetrahydropalmatine was Y = 6.54 × 10 ~ 6X + 1.55 × 10 ~ 4 (r = 0.9996). The results showed that the tetrahydropalmatine injection volume in the range of 0.02001 ~ 0.2001μg good linear relationship, HPLC measured Shijiazhuangbucao wild baicalin, Codonophor glycosides, tetrahydropalmatine through isolated rat skin 12h transdermal Rates were 20.06%, 19.23%, 28.81% respectively. Conclusion The transdermal permeation behavior of SD rats is a passive diffusion behavior in vitro, and the transdermal flux increases with the increase of drug concentration.
