建立了粮食中黄曲霉毒素B1、B2、G1、G2的免疫亲和柱净化-光化学柱后衍生高效液相色谱荧光检测法。样品经甲醇-水提取,免疫亲和柱净化,高效液相色谱分离,光化学柱后衍生,荧光检测器测定。结果表明,黄曲霉毒素B1、B2、G1、G2的检出限分别为0.50、0.25、0.50、0.25μg/kg,回收率为67.2%~91.7%,RSD小于10%。该方法快速、准确、灵敏度高、重现性好,能满足我国对粮食中黄曲霉毒素限量的检测要求。
An immunoaffinity column purification of aflatoxins B1, B2, G1 and G2 in food was established by fluorescence post column derivatization with high performance liquid chromatography. The samples were extracted by methanol - water, purified by immunoaffinity column, separated by high performance liquid chromatography, post - column derivatized by photochemical method and detected by fluorescence detector. The results showed that the detection limits of aflatoxins B1, B2, G1 and G2 were 0.50, 0.25, 0.50 and 0.25 μg / kg, respectively. The recoveries were 67.2% ~ 91.7% with RSD less than 10%. The method is rapid, accurate, sensitive and reproducible. It can meet the detection requirements of aflatoxin in food in our country.
