目的研究红景天苷对高糖条件下培养的胰岛β细胞功能的影响。方法用30m M葡萄糖培养INS-1胰岛细胞,加入不同浓度的红景天苷(0.1μΜ、1.0μΜ和10μΜ)进行干预,并用5.6 m M浓度的葡萄糖作为阴性对照。通过MTT检测INS-1细胞活性、Real time PCR检测胰岛素、TRX、TXNIP mRNA表达变化;用酶联免疫法检测培养上清胰岛素分泌量;使用活性氧检测试剂盒检测细胞内自由氧生成水平。结果体外诱导条件下,高浓度葡萄糖通过上调TXNIP mRNA以及下调TRX mRNA表达,使INS-1细胞内ROS升高,降低胰岛素的分泌;而红景天苷通过下调TXNIP mRNA、上调TRX mRNA水平,维持细胞内ROS的低水平,提高胰岛素的分泌。结论红景天苷对胰岛β细胞的糖毒性损坏具有保护作用。
Objective To study the effect of salidroside on the function of islet β cells cultured in high glucose. Methods INS-1 islet cells were cultured with 30m M glucose, and different concentrations of salidroside (0.1μM, 1.0μM and 10μM) were used for intervention, and 5.6m M glucose was used as a negative control. The activity of INS-1 cells was detected by MTT, the mRNA expression of insulin, TRX and TXNIP were detected by Real time PCR. The secretion of insulin in supernatant was detected by enzyme-linked immunosorbent assay (ELISA). Results Under high concentration of glucose, high concentration of glucose upregulated TXNIP mRNA and down-regulated the expression of TRX mRNA in vitro, which led to the increase of ROS in INS-1 cells and the decrease of insulin secretion. However, salidroside upregulated TRX mRNA by down-regulating TXNIP mRNA Low levels of intracellular ROS, increase insulin secretion. Conclusion Salidroside has a protective effect on the isotoxicity of pancreatic islet β cells.
